How to Take Foundayo
Foundayo (orforglipron) is taken as a once-daily oral tablet. It can be taken at any time of day, with or without food, making it a convenient option for people looking for an oral GLP-1 weight loss treatment.
- Take Foundayo once daily, at approximately the same time each day if possible.
- Foundayo can be taken with or without food and does not require you to take it on an empty stomach.
- There are no specific water restrictions when taking Foundayo.
- Swallow the tablet whole. Do not break, crush or chew the tablet.
- Do not take more than one Foundayo tablet per day.
- If you miss a dose, take it as soon as possible. Do not take two doses on the same day.
- If you have missed Foundayo for 7 days or more, contact your prescriber for advice on how to restart treatment.
Foundayo Dose and Treatment Schedule
Foundayo is available in 0.8 mg, 2.5 mg, 5.5 mg, 9 mg, 14.5 mg and 17.2 mg tablet strengths. The maximum recommended dose is 17.2 mg once daily.
Treatment is gradually increased to help your body adjust to Foundayo and may help reduce gastrointestinal side effects. Your prescriber will determine the most appropriate dose based on your response to treatment, tolerability and individual circumstances.
The standard dose escalation schedule is:
- First 30 days: 0.8 mg once daily
- Days 31–60: 2.5 mg once daily
- Days 61–90: 5.5 mg once daily
- From day 91 onwards: Your prescriber may increase your dose to 9 mg, 14.5 mg or 17.2 mg once daily, depending on your response and tolerability.
Each dose should generally be maintained for at least 30 days before considering an increase. Your prescriber may recommend staying at your current Foundayo dose for longer if this is appropriate for you. The aim is to find the dose that provides an appropriate balance between treatment response and tolerability.
You may experience more noticeable Foundayo side effects, particularly gastrointestinal symptoms such as nausea, vomiting, diarrhoea, constipation or abdominal discomfort, when starting treatment or increasing your dose. Tell your prescriber if side effects are troublesome or persistent.
When will Foundayo start working?
Weight loss with Foundayo develops over time and individual results vary. Clinical studies have demonstrated weight loss at different doses of orforglipron when used alongside a reduced-calorie diet and increased physical activity. Your prescriber will monitor your progress and determine whether your current Foundayo dose remains appropriate.
Switching to Foundayo from Another GLP-1 Treatment
If you are currently taking Wegovy (semaglutide), Mounjaro (tirzepatide) or another GLP-1 receptor agonist, do not change your treatment or start Foundayo without speaking to your prescriber.
There is no simple dose-for-dose conversion between Foundayo and other GLP-1 medicines. The appropriate approach to switching will depend on your current medication and dose, treatment history, medical circumstances and how well you have tolerated your existing treatment.
Your prescriber will assess your current medication and determine whether switching to Foundayo (orforglipron) is appropriate and, if so, which Foundayo dose and treatment schedule should be prescribed.
Foundayo should not be taken at the same time as another GLP-1 receptor agonist.

Foundayo Side Effects
Common side effects may include:
- Nausea

Gastrointestinal side effects such as nausea, diarrhoea, constipation, vomiting and abdominal discomfort are among the most commonly reported effects with Foundayo (orforglipron). These effects may be more noticeable when starting treatment or increasing your dose and often become less troublesome as your body adjusts to treatment.
Serious side effects to be aware of:
- Serious allergic reactions: Seek immediate medical attention if you experience swelling of the face, lips, tongue or throat, difficulty breathing, severe dizziness or other symptoms of a serious allergic reaction.
- Acute pancreatitis: Severe or persistent abdominal pain, particularly if it spreads to the back, with or without vomiting, may be a sign of pancreatitis and requires urgent medical attention.
- Gallbladder problems: Foundayo may increase the risk of gallstones and inflammation of the gallbladder. Seek medical advice if you develop severe or persistent abdominal pain, particularly in the upper abdomen.
- Dehydration and kidney problems: Persistent vomiting or diarrhoea can cause dehydration and, in some cases, acute kidney injury. Speak to your healthcare professional if you are unable to maintain adequate fluid intake.
- Low blood sugar (hypoglycaemia): The risk may be increased when Foundayo is used alongside insulin or certain other diabetes medicines, such as sulfonylureas.
- Diabetic retinopathy complications: If you have diabetic retinopathy, tell your healthcare professional before starting Foundayo, particularly if you experience changes in your vision.
Foundayo also carries a warning concerning thyroid C-cell tumours, including medullary thyroid carcinoma (MTC). Foundayo should not be used if you have a personal or family history of MTC or Multiple Endocrine Neoplasia syndrome type 2 (MEN 2).
Most side effects associated with Foundayo weight loss treatment are gastrointestinal and may improve as your body becomes accustomed to the medication. Gradual dose escalation is used to help improve tolerability. Your prescriber may recommend staying at your current dose for longer if you are experiencing troublesome side effects.

Foundayo Warnings & Safety Information
Who should not take Foundayo?
Do not use Foundayo (orforglipron) if you have a personal or family history of medullary thyroid carcinoma (MTC) or Multiple Endocrine Neoplasia syndrome type 2 (MEN 2), or if you have previously experienced a serious allergic reaction to orforglipron or any of its ingredients.
Pancreatitis
Tell your prescriber if you have previously had pancreatitis. Seek urgent medical attention if you develop severe or persistent abdominal pain, particularly if it spreads to your back.
Stomach and digestive problems
Foundayo can cause gastrointestinal side effects such as nausea, vomiting, diarrhoea, constipation and abdominal discomfort. It is not recommended for people with severe gastroparesis or severe problems with stomach emptying.
Dehydration and kidney problems
Severe vomiting or diarrhoea can cause dehydration and, in some cases, kidney problems. Contact your healthcare professional if you experience persistent gastrointestinal symptoms or have difficulty maintaining your fluid intake.
Gallbladder problems
Foundayo may increase the risk of gallstones and inflammation of the gallbladder. Seek medical advice if you experience severe or persistent abdominal pain.
Low blood sugar
Foundayo may increase the risk of hypoglycaemia, particularly when used with insulin or certain diabetes medicines. Tell your prescriber if you take medication for diabetes.
Vision problems
If you have diabetic retinopathy, tell your healthcare professional before starting Foundayo. Contact your healthcare provider if you experience changes in your vision.
Other medicines
Foundayo can delay stomach emptying and may affect the absorption of some oral medicines. Tell your prescriber about all medicines, supplements and vitamins you take.
Pregnancy and breastfeeding
Do not use Foundayo during pregnancy or if you are planning to become pregnant. Breastfeeding is not recommended during treatment. Speak to your healthcare professional if you become pregnant while taking Foundayo.
Surgery or anaesthesia
Tell your healthcare professional if you are scheduled for surgery, general anaesthesia or deep sedation, as Foundayo may delay stomach emptying.
Children and young people
Foundayo is intended for eligible adults. Its safety and effectiveness have not been established in people under 18.
When to seek urgent help
Seek immediate medical attention if you experience difficulty breathing, swelling of the face, lips, tongue or throat, severe or persistent abdominal pain, or other symptoms of a serious allergic reaction or pancreatitis.
